While the sequence of amino acids is a critical determinant of how a protein folds and behaves, some proteins play by a different ruleset. These proteins have &#8220;composition-driven activities&#8221;, where the amino acid composition is a stronger determinant of the activity than the linear sequence of amino acids. This review discusses known examples of composition-driven activities, as well as experimental criteria for classifying activities as composition-driven.
